Two different bait vectors, pLexA-N and pLexA-C, allow high-level, constitutive expression of bait proteins fused either to the C-terminus or to the N-terminus of the bacterial pLexA-N domain. Vectors carry a kanamycin marker to facilitate plasmid rescue and can be combined with most commercially available cDNA libraries.
The pGAD-HA library (prey) vector is a constitutive, high-level expression vector suitable for the DUALhybrid and most other yeast two-hybrid systems.
The highly sensitive yeast host strain NMY1 features three reporter genes for superior screening stringency. Colonies likely to contain an interacting pair of proteins will grow on media lacking histidine and adenine. In addition, putative positive colonies expressing ß–galactosidase will turn blue when incubated with X-gal or a similar substrate due to the presence of the lacZ reporter gene in the NMY1 strain.
The bridging vector pY3H allows expression of a third binding partner for yeast three-hybrid studies.
