HTX Beta-galactosidase Assay Kit (Cat # P01002)

 

 

In a typical yeast two-hybrid or DUALmembrane screen, dozens or even hundreds of putative interactors are often identified. Commonly used assays to detect beta-galactosidase activity, such as the filter lift assay or the ONPG assay, are either not quantitative or very cumbersome to use. In contrast, the HTX kit yields semi-quantitative measurements with minimal handling: less than 30 minutes for 96 clones!

Yeast cultures are grown in a user-supplied 96 deep-well block until they have reached the late exponential phase and pelleted by centrifugation. Pellets are lysed by addition of the one-step lysis/substrate buffer and transferred to a user-supplied clear-bottom 96 well plate. Color development over time is monitored by eye and recorded using either a regular flatbed scanner or a spectrophotometer.
